Laboratory and field assessment of molluscicidal activity of B-2 against Oncomelania hupensis, the vector snail of schistosomiasis in China

Molluscicidal activity of B-2 (sodium 2,5-dichloro-4-bromophenol; called as Phebrol and registered in WHO as OMS 3012) was evaluated in a laboratory and the field trials were performed in two different localities in Yueyang city, China, for control of Oncomelania hupensis. B-2 was effective against O. hupensis both in the laboratory and in the field. A dosage of 50 g/m2 in 10% granular form or 20 ml/m2 in 25% liquid form of B-2 would be recommendable as a standard mollusciciding dose for control of O. hupensis.     

Cloning, sequencing and functional expression of a cDNA encoding porcine pancreatic preprocarboxypeptidase A1.

A full-length cDNA clone coding for porcine pancreatic preprocarboxypeptidase A1 (prePCPA1) was isolated from a cDNA library. The open reading frame (ORF) of the nucleotide sequence was 1260 nt in length and encoded a protein of 419 amino acids (aa). The cDNA included a short signal peptide of 16 aa and a 94 aa-long activation segment. The calculated molecular mass of the mature proenzyme was 45561 Da, in accordance with that of the purified porcine pancreatic PCPA1. The deduced aa sequence of the corresponding enzyme differed from that predicted by the three-dimensional structure by 40 aa, and showed 85% identity and 55% identity to that of procarboxypeptidases A1 and A2, respectively. Moreover the sequence was identical to that of several independent cDNA clones, suggesting that it is the major transcribed gene. No evidence for a second variant was observed in the cDNA library and PCPA2 is apparently absent from the porcine pancreas. The cDNA was expressed in Saccharomyces cerevisiae under the control of the yeast triose phosphate isomerase promoter. The signal peptide of the PCPA protein efficiently directed its secretion into the culture medium (1.5 mg.L-1) as a protein of the predicted size. The recombinant proenzyme was analyzed by immunological and enzymological methods. Its activation behavior was comparable with that of the native form and led to a 35-kDa active enzyme.     

Division of Labour among Workers in the Termite,Reticulitermes fukienensis (Isoptera: Rhinotermitidae)

To investigate division of labour in Reticulitermes fukienensis, worker and larval subgroups were established in laboratory cultures and task performance patterns of different-sized workers and larvae were recorded. Five categories of subgroups were established based on head width differences: small larvae (SL); large larvae (LL); small workers (SW); medium workers (MW); and large workers (LW). Subgroups were compared for their ability to carry out the following tasks: tunnel construction; covered gallery construction; gallery repair; and feeding. Larval subgroups were found to carry out none of the tasks investigated. They did not feed or build covered galleries or underground tunnels. Temporal polyethism occurred among workers of different size groups. Covered foraging galleries were exclusively built and tunnels were predominantly built by older workers (i.e. MW and especially LW). Remarkably, most SW were apparently unable to burrow into the soil, something that all LW did within 3 d and 95% of MW did within 5 d. Surprisingly, LW ate 8.5 and 65.5 times as much food (i.e. filter paper) as MW and SW, respectively, although their average body weights were, respectively, only approximately two and four times that of MW and SW. MW carried out all four tasks that LW performed, although MW were typically less efficient. This contrasts with the hypothesis that different instars of termites should carry out different non-overlapping tasks. Large workers carried out most of all tasks compared with other worker sizes. The only exception was for gallery repair where repair rate by LW did not differ significantly from MW. The predominance of tasks carried out by larger (i.e. older) workers indicates a possible new pattern for division of labour in these lower termites. This contrasts with the pattern of organization of division of labour in the social Hymenoptera.

Discovery of Iliestheria (Crustacea: spinicaudatan) from the Lower Jurassic Sangonghe Formation in Junggar Basin,northwestern China

Iliestheria Li and Shen, 1995, was erected based on specimens of two co-occurring species from the Lower Jurassic Badaowan Formation in Nilka, Xinjiang Uygur Autonomous Region, northwestern China. Since then Iliestheria has not been reported in any other locality, partly because clam shrimp from the Lower Jurassic in western China have been reported with studies only based on the outline of the carapace, most of them as Euestheria or Palaeolimnadia components. Here we report a new species of Iliestheria from the Pliensbachian–Toarcian Sangonghe Formation, based on scanning electron microscopic characteristics of the carapace morphology. We also revise the diagnosis of the genus based on re-examination using SEM microscopy of the two species from the Badaowan Formation, to include spinicaudatans characterized by growth bands ornamented by radial lirae intercalated with 2–4 columns of puncta; radial lirae sporadically curved and branched to form fine reticulation on the growth bands in the anterior and upper parts of the carapace. This study indicates that the stratigraphic range of Iliestheria extends to late Early Jurassic.
